C57BL/6 Mouse Embryonic Stem Cells

When C57BL/6 was implanted into c2J blastocyst, the yield of hair color chimera was high; However, the number of chimeras produced by implantation of FVB blastocyst is small. ES cells can enter the germ line of the blastocyst of the donor host, FVB/NJ (FVB) and the homologous mutant strain C57BL/6-Tyrc-2J (c2J). CF-1 MEF (SCSP-105R) should be used as feeder layer cells during culture. It was put into storage after identification without mycoplasma.

MES complete culture solution formula (600 ml): DMEM (Invitrogen 12430) 497 ml ES grade FBS (biochrom S4115) 90 ml Glutamax (Invitrogen 35050) 6 ml NEAA (Invitrogen 11140) 6 ml LIF (Millipore ESG1107) 60 μ L (final concentration is 1000U/ml) β- Mer (Invitrogen 21985) 1.5 ml

Gas phase: air, 95%; Carbon dioxide, 5%. Temperature: 37 ℃.

Freezing conditions: basic medium+5% DMSO+20% FBS (SD0200)

Complete cryopreservation solution: non-programmed cooling cell cryopreservation solution (low serum) (SD0001)
