Purinomycin solution

For the screening and maintenance of cell stable strains (overexpressed or silenced cell lentivirus stable strains and LUS.GFP.RFP cells), the concentration of this product is 1mg/ml, please add it according to the equivalent concentration after cell screening

Screening of working concentration of Puromycin: 1. Cells were laid on 24-well plates 24h in advance, and the number of cells was adjusted according to the growth state and size of cells. After 24h, the cell density was about 70%; 2. After 24h of cell growth, puromycin with different final concentrations was added, as shown in the figure; 3. After 48 hours of culture, the puro group with the lowest concentration of all killed cells was observed under the microscope as the working concentration

After 48-72 hours of lentivirus infection (70-80% fusion degree), the cells were continued to culture in culture medium containing appropriate concentration of Puromycin. After 48 hours of screening, all the empty cells plus puro group died, indicating successful screening.
