Testicular Epithelial Cells Of 15P-1 Mice

15P-1 cells are derived from the testicular cells of transgenic mice that express large T protein of polytumor virus (PyLT) in spermatogenic epithelium. This cell shows the characteristics of Sertoli cells, including the transcription of Wilms tumor (WT1) and Steel gene. It was reported that 15P-1 culture showed phagocytic activity, which was proved by the endocytosis of latex particles and the elimination of dead cells. The interaction with germ cells enhanced the phagocytic activity of 15P-1 cells. When co-cultured with diploid germ cells in the prophase of meiosis, the cells support spermatogonia to differentiate into haploid sperm expressing protamine (Prm-1) gene after meiosis and meiosis. Testicular cells were taken from immature 9-day-old animals before the first meiosis and co-cultured with 15P-1 cells to produce tetrads with round sperm morphology and initiate protamine transcription. 15P-1 cells can produce substances sensitive to protease and with broad-spectrum antibacterial activity. In the presence of components rich in round sperm and nerve growth factor, the antibacterial activity of 15P-1 medium increased by 10 to 50 times. KL protein (ligand of Kit receptor) was positive in Sertoli cells and 15P-1 cells. It was put into storage after identification without mycoplasma.
