PA317 Mouse Embryonic Fibroblast

The PA317 cell line was derived from TK-NIH/3T3 cells and was constructed by co-transfection of the packaging structure DNA (pPAM3) in pBR322 and the herpes simplex virus thymidine kinase gene in pBR322. Retroviral vectors are introduced into these cells through infection or transfection to generate biphilic vector particles that can infect many mammalian cells. The virus particles generated by this cell have been successfully used to introduce genes into human body. The percentage of PA317 cells capable of packaging retroviral vectors decreased with passage, possibly due to the loss of transferred DNA. Cells that retain the packaging function can be selected in the medium containing 0.03 mM hypoxanthine, 0.001 mM methotrexate and 0.02 mM thymidine for a short time (about 5 days). It was put into storage after identification without mycoplasma.
