AC16 human cardiac cell line is a proliferative cell line, which is derived from the fusion of primary cells from adult ventricular myocardial tissue and SV40 transformed uridine deficient human fibroblasts, lacking mitochondrial DNA (1,2). After the unfused fibroblasts were removed in urine-free medium, the fusion cells were further subcloned, and then the existence of SV40 large T-ag was screened, β- Myosin heavy chain( β MHC) and connexin 43 (CX-43) (1). The proliferation and differentiation of AC16 cells may be controlled by changing the culture conditions and inhibiting the expression of sv40t-Ag. These cells can study the gene expression and function of the heart under normal development and pathological conditions at the cellular, organelle and molecular levels; Cells can be cultured for at least 10 generations after initial thawing without significantly affecting the expression and function of cell markers.
