DLD-1 Human Colorectal Adenocarcinoma Epithelial Cells

DLD-1 is one of two colorectal adenocarcinoma cell lines isolated by D.L. Dexter and colleagues from 1977-1979. DNA fingerprinting and karyotype analysis in ATCC and other places showed that this cell line was similar to HCT-15 (CCL-225), indicating that the two were different clones from the same person. Their genetic origin can be confirmed by DNA fingerprinting, but karyotype analysis shows that they lack consistent changes in chromosome markers or numbers. The cell's CSA p is negative (CSA p -). The expression of p53 antigen in DLD-1 cells was positive (p53 antigen produced a C ->T point mutation leading to Ser ->Phe at 241). Keratin immunoperoxidase staining was positive. The expression of oncogenes c-myc, K-ras, H-ras, N-ras, myb, sis and fos was positive. The expression of oncogene N-myc was not detected. Expression of tumor specific nuclear matrix proteins CC-2, CC-3, CC-4, CC-5 and CC-6. The culture submitted to ATCC in 1979 was of unknown generation and contaminated with mycoplasma. After 12 weeks of combined culture with multiple antibiotics. After treatment, Hoechst staining and standard culture method were used for detection every week. After that, all tests were negative without antibiotic culture for 11 consecutive months.
