HPAC Human Pancreatic Cancer Cells

HPAC is a kind of pancreatic adenocarcinoma epithelial cell line. In 1985, a nude mouse xenograft tumor with primary tumor was removed from the pancreatic head of a woman with moderate to well-differentiated ductal pancreatic adenocarcinoma. HPAC cells were keratin positive, vimentin and chromaffin A negative. The cells formed tumor at the inoculation site of thymus-free nude mice, which was similar to the origin of tumor in histology. The colony forming efficiency of cells on the culture plate was 64% and that on agarose was 3.2%. Insulin, insulin-like growth factor I (IGF-I), epidermal growth factor (EGF), and transforming growth factor alpha (TGF - alpha) can stimulate HPAC proliferation. Dexamethasone and other glucocorticoids can inhibit cell growth. HPAC cells contain a large number of tumor related antigens CEA, ca125 and ca19-9. In the process of culture, HPAC cells formed heterogeneous monolayer polar epithelial cells. HPAC is the first reported human pancreatic adenocarcinoma cell line expressing functional glucocorticoid receptor. Immunohistochemical staining showed that HPAC cells expressed DU-PAN-2, a marker of pancreatic duct epithelium, and antigens recognized by monoclonal antibodies HMFG1 and AUA1.
