The THLE-3 cell line originates from primary normal hepatocytes in the left lobe of the human liver and achieves immortality through SV40 large T antigen infection. The specific process involves introducing a retroviral vector containing the Bgl I-Hpa I fragment containing SV40 T antigen into the concurrent philophilic packaging cell line PA317 to produce the virus, which then infects normal hepatocytes and establishes the THLE-3 cell line. THLE-2 and THLE-3 cell lines originated from primary hepatocytes in the same left lobe of normal liver, using the same retroviral vector (Bgl I-Hpa I fragment containing SV40 T antigen), and were produced by PA317-packaged cell lines to infect hepatocytes and thus establish the above cell lines. THLE-3 cells express phenotypic features of normal adult hepatic epithelial cells, such as cytokeratin 18 and albumin, and cytokeratin 19 is also expressed in high-pass cells during the log-growth phase. This cell line has a nearly diploid karyotype, does not express alpha-fetoprotein, and does not form tumors in nude mice without thymos. THLE-3 cells retain enzymes related to chemical tumor metabolism, including cytochrome P450 pathways, epoxide hydrolase, NADPH cytochrome P450 reductase, superoxide dismutase, catalase, glutathione S-transferases, and glutathione peroxidase can be used to absorb benzo[a]pyrene, N-nitrosodimethylamine, and aflatoxin B1) and other chemical carcinogens are metabolized into the final carcinogenic metabolites. These metabolites can bind to DNA, making THLE-3 cells an in vitro model for studying drug toxicology and the pathogenic and pathological studies of human hepatocellular carcinoma.
