NR8383 [AgC11x3A, NR8383.1] Rat Alveolar Macrophages

NR8383 (normal rats, August 3, 1983) is derived from normal rat alveolar macrophages during lung lavage. The cells were cultured in the presence of continuous culture medium of gerbil lung cells for about 8-9 months. Subsequently, exogenous growth factors are no longer needed. NR8383 cells were cloned and subcloned from single cells by limited dilution method, and subcloned with soft agar three times. The cells showed the characteristics of macrophages, phagocytosis of yeast polysaccharide and verdigris, non-specific lipase activity, Fc receptor, oxidative degradation; Secrets IL-1, TNF beta and IL-6, which can repeatedly respond to exogenous growth factors. NR8383 cells respond to bleomycin and secrete TGF beta precursor. Under the stimulation of bleomycin, the expression of TGF beta mRNA also increased. Cells are sensitive to endotoxin. LPS levels of 1-10 g/ml inhibited proliferation by up to 50%. Even at the level of 0.001 mg/ml, LPS inhibition is non-toxic and reversible in the subsequent process. NR8383 cell line provides a uniform source of highly responsive alveolar macrophages, which can be used to study the related activities of giant cell in vitro. This cell grows in semi-suspension.
