What if the extracted cells are impure? Follow the following method for magnetic bead screening to easily solve the problem of impure cells.

1. Prepare magnetic beads and other necessary reagents, and the storage temperature for all sorting reagents should be 2-8C. Low or high temperatures can affect the expiration date, thereby affecting the effectiveness of the product;


2. Preheat the reagents in advance, and all reagents and samples should be balanced to room temperature (15-25C) before use;


3. All operations should be carried out in the ultra clean table or biosafety cabinet, with room temperature controlled at 15-20 ℃;


4. Manual sample processing requires high laboratory operating techniques and meticulous operations. When using BeadRetriever, the operation of transferring the sample to the test tube should be carried out in an area at least 1 meter away from the prepared sample;


5. The magnetic beads must be shaken and mixed evenly before use;


6. Dilution can be used when handling extremely viscous or high fat content samples. It is recommended to use a specific washing buffer (PBS Tween) to dilute the sample;


7. Use a 360 "rotary mixer to incubate magnetic beads. Suggest rotating from top to bottom several times;


8. Incubate at room temperature for 10 minutes. Excessive incubation only slightly improves the recovery ability of target cells or molecules, but increases the possibility of non-specific binding;


9. Wash thoroughly and perform the prescribed number of times;


10. Mix the liquid slightly, instead of shaking it well. Before applying a magnetic field, ensure that the magnetic beads are properly added to the rolling liquid;


11. When concentrating the magnetic beads in a small ball, perform 1/2 of each rotation using a 90 ° rock n roll motion at a specific time;


12. Carefully remove the suspension. Manually suck the liquid out of the test tube instead of empty suction. After suctioning magnetic beads from the sample tube, as these liquids need to be discarded, it can cause the loss of target cells or molecules;


13. Additional washing steps can reduce background interference, but also peak the recovery efficiency of target cells;


14. If background pollution is considered, the centrifuge tube can be replaced after each washing step. Many microorganisms can adhere to the tube wall, and replacement can reduce liposome contamination.
