MM.1R Human Multiple Myeloma Cells

The mother cell line MM. 1 of this cell line was established from the peripheral blood of a patient with multiple myeloma who was resistant to steroid therapy. MM.1R is resistant to dexamethasone. The related cell line MM. 1S was also isolated from MM. 1, but was sensitive to dexamethasone. The cells need about two weeks to resume normal growth after resuscitation. It was identified as free of mycoplasma/STR and put into storage.

RPMI Medium 1640 (Invitrogen 11875093) 87 ml

FBS (Gibco) 10 ml

Glutamax（invitrogen 35050061） 1 ml

Non-essential Amino Acids, 100′ (Invitrogen 11140050) 1 ml

Sodium Pyruvate 100 mM Solution（invitrogen 11360070） 1 ml

Gas phase: air, 95%; Carbon dioxide, 5%. Temperature: 37 ℃.

Reference generation ratio: 1:2-1:4

Reference generation cycle: 3-5 days

Reference fluid change frequency: 2-3 days

Freezing solution formula: complete culture solution 90%, DMSO 10%

[Precautions]

The culture process of this cell line contains both monolayer cells with slight adhesion and suspension cells. The culture can be maintained by adding fresh medium. During passage, the adherent cells were scraped into the culture medium containing floating cells by cell scraping, the cells were collected by centrifugation, the cell particles were resuspended in the fresh culture medium and inoculated into a new bottle. Note: The cell density should not be too large, otherwise the over-fused adherent cells begin to form rosette structure with necrotic center.
