Products Description

In August 1991, Kristina Vintersten from the European Molecular Biology Laboratory (EMBL) in Heidelberg, Germany, successfully established R1 cells using a 3.5-day blastocyst from the 129X1×129S1 line; R1/E cells are subclones of R1. This cell line has been proven to have reproductive ability, with the F1 generation having a single mouse gray coat color, and the F2 generation having two coat color genes separated. This separation may result in several coat colors, including white, light brown, and black, depending on the genetic background of the parent of the hereditary chimera. R1 pluripotency was initially detected by tetraploid embryos, though diploid embryos can also be used. However, when tetraploid aggregate detection was performed, about 20% of subclones from the 14th generation showed the original developmental potential of R1. Animals derived from R1 are fertile once they reach adulthood. Injecting genetically modified R1 cells into C57 blastocysts or aggregating them with CD-1 or outbred ICR 8-cell stage embryos can produce efficient germline transfer. Over 90% of knockout clones enter the germline through aggregated chimeras (n> 60). Each batch passed mycoplasma testing in the database and was negative.
During culture, Kunming MEF should be used as the culture layer cells.
