MC3T3-E1 Subclone 24 cells and their subclones are excellent models for in vitro studies of osteoblast differentiation, especially regarding the role of extracellular matrix (ECM) signaling pathways. Their behavior is similar to that of primary cranial osteoblasts, capable of synthesizing and secreting bone matrix proteins such as alkaline phosphatase and osteocalcin, and responsive to various bone metabolism regulators. After growing in medium containing ascorbic acid, subclones were selected based on high or low osteoblast differentiation and mineralization. After growing in ascorbic acid and 3-4 mM inorganic phosphate, the MC3T3-E1 Subclone 4 and MC3T3-E1 Subclone 14 cell lines exhibited high levels of osteoblast differentiation, forming well-mineralized extracellular matrix (ECM) after 10 days. The MC3T3-E1 Subclone 24 and MC3T3-E1 Subclone 30 cell lines exhibited poor osteoblast differentiation after growth in ascorbic acid and did not form ECM. Therefore, MC3T3-E1 Subclone 24 can be used as a negative control for the MC3T3-E1 Subclone 4 and MC3T3-E1 Subclone 14 cell lines. It holds significant value for studying in vitro osteoblast differentiation, especially in extracellular matrix (ECM) signaling. In addition, the MC3T3-E1 Subclone 24 cell line is also widely used in bone biology and calcification research, especially in studying osteoblast differentiation and mineralization.
