RWPE-1 Human Normal Prostate Epithelial Cells

The epithelial cells in the surrounding area of normal prostate tissue section of a 54-year-old white man were transformed with a single copy of human papillomavirus 18 (HPV-18) to establish RWPE-1. It is reported that RWPE-1 cells form acini and secrete PSA into the culture medium under the stimulation of androgen when cultured in three-dimensional matrix. When mixed with matrigel or stroma cells to inject male nude mice, RWPE-1 cells can also form glandular cells and produce PSA. Cells derived from RWPE-1 were transfected with Ki-ras gene with Kirstin mouse sarcoma virus (Ki-MuSV) to establish tumorigenic RWPE-2 cells and RWPE2-W99 cells. In addition, RWPE-1 was treated with N-methyl-N-nitrourea (MNU) to establish a series of tumorigenic cell lines simulating different stages of prostate cancer progression. They are WPE1-NA22, WPE1-NB14, WPE1-NB11 and WPE1-NB26 cells. It is reported that RWPE-1 cells are negative for hepatitis B virus, hepatitis C virus and human immunodeficiency virus after detection. PCR confirmed that the HPV DNA sequence of the cell line was positive. It was identified as free of mycoplasma/STR and put into storage.
