0.25% Trypsin Digestion Solution/EDTA

Trypsin is produced by the pancreas and secreted into the small intestine by inactive trypsinogen. The intestinal peptidase in the small intestine will activate the trypsinogen to form trypsin. The characteristic of trypsin is that the activated trypsin can continue to activate more trypsinogen. This process is called autocatalysis. Trypsin works in the small intestine. It will hydrolyze protein into peptides, and then decompose into amino acids. The optimum temperature is about 37 ℃.

EDTA is ethylenediamine tetraacetic acid, a metal chelating agent. It is generally used in combination with trypsin. The reason is that calcium, magnesium and other metal ions will reduce the activity of trypsin, so EDTA should be added when using trypsin digestive solution. It can chelate these ions and eliminate the inhibition of trypsin

The trypsin digestion solution (including EDTA) is composed of 0.25% trypsin and 0.02% EDTA, and is filtered. This reagent can be directly used for the digestion of cultured cells or some tissues. Usually, most adherent cells can be digested in about 2 minutes at room temperature.

Three. The time for trypsin digestion solution to digest cells should not be too long, otherwise the growth of cells will be poor after planking.

Four. It is best to use cells in logarithmic growth phase for passage digestion.

Five. Control the digestion time of pancreatin, and remember that excessive digestion will affect the effect of cell resuscitation.

Six. During the process of cell resuspension, please be gentle to avoid damaging the cells, but at the same time, it must also be fully resuspended so that the cells will not form a cluster during the recovery.
