The cells were sampled from a 39-year-old male patient and tested by STR/ Mycoplasma-free in this library. Nude mice were inoculated with human hepatocellular carcinoma cell line MHCC97-H, lung metastasis screening was performed three times, and lung metastasis was constructed into a highly spontaneous lung metastasis cell line after subcutaneous inoculation.
This is the labeled Luc cell line. If the fluorescence expression is not strong, the floating cells can be discarded after 48 hours of purinol screening with 2ug/ml, and replaced with a normal culture system, which can improve the fluorescence expression of cells. Luciferase is an oxidase that gives off light in the presence of a substrate (D-luciferin) in organisms, a process called bioluminescence. The most common luciferase genes come from a family of light-producing enzymes called firefly luciferase 1 and 2.
In preclinical research, live imaging of small animals plays a vital role in visualizing physiological processes, disease progression, and therapeutic development. Luciferase (luc) -activated cell lines via bioluminescence imaging (BLI) 1,2 provide a simple, high-throughput, and robust method for quantitative assessment of tumor burden and tumor response to treatment in test animals.
Luc cell labeling has several advantages, such as:
(1) Non-invasive real-time whole-body tumor monitoring and imaging
(2) Ongoing assessment of tumor progression and treatment response in the same animal
(3) Transfer assessment
(4) Reduce the need for animal sacrifice
