The VK2/E6E7 cell line is a human vaginal epithelial immortalized cell line established in 1996 by D. Anderson, R.N. Fichorova, and J.G. Rheinwald. The primary cells were taken from the normal vaginal mucosa tissue of a 32-year-old premenopausal woman with factor endometriosis who underwent hysterectomy. During strainmaking, the third-generation primary cells were infected with the third-generation primary cell with the retroviral vector LXSN-16E6E7 containing the HPV-16 E6/E7 gene, and clones were screened in G418-containing medium to achieve immortality. These cells continuously expressed the cells keratin CK8, CK10, CK13, CK18, CK19, as well as vaginal epithelial-associated proteins such as M-CSF, TGF-β1, IL-8, PGE2, SLPI, and pIgR. VK2/E6E7 cells can serve as normal vaginal epithelial cell models, providing an effective and reproducible in vitro model for studying cervicovaginal physiology and infection, as well as testing drug formulations for vaginal applications. Due to its clear origin, stable growth, and retention of most primary vaginal epithelial functions, VK2/E6E7 has become one of the "gold standards" for in vitro genital tract evaluation models.
