H9c2(2-1) Cas9-ZsGreen-Luc

The H9c2(2-1) Cas9-ZsGreen-Luc cells were generated by slow viral infection to achieve stable co-expression of HBLV-Cas9-Puro and HBLV-ZsGreen-Luc in these cells. Monoclonal lines were selected following screening with 2.5 μg/mL puromycin. Validation through PCR, Western blotting, and luciferase reporter assays confirmed that the H9c2(2-1) Cas9-ZsGreen-Luc cells stably express the Cas9 protein while simultaneously carrying the green fluorescent protein ZsGreen and luciferase (Luc) markers, enabling direct and more efficient utilization for genomic DNA editing via CRISPR technology.
