HK-2 Human Renal Epithelial Cells

It is reported that human renal proximal convoluted tubule cell HK-2 contains a recombinant retroviral vector pLXSN 16 E6/E7 containing HPV-16 E6/E7 gene. Although pLXSN 16 E6/E7 is resistant to neomycin, G418 cannot be used to isolate the transduced clone. The cell line can be separated into single cells based on Southern and FISH analysis. The E6/E7 gene was detected in the HK-2 genome by PCR. The cells retained well differentiated PTCs phenotype. Alkaline phosphatase γ Glutamyl transpeptidase, leucine aminopeptidase, acid phosphatase, cytokeratin α 3、 β The expression of integrin and fibronectin was positive. The expression of factor VIII associated antigen, 6.19 antigen and CALLA endopeptidase was negative. HK-2 cells retain the functional characteristics of proximal renal tubular epithelial cells, such as Na+dependent/phlorizin-sensitive glucose transport and adenylate cyclase response to parathyroid gland, but are not sensitive to antidiuretic hormone. This cell has the ability to generate and store glycogen. HK-2 cells grew adherent to the wall, did not grow on methylcellulose and soft agar, and could not grow in suspension.
