The Alizarin Red S staining kit is primarily used for osteogenic differentiation identification: it detects whether mesenchymal stem cells, among others, have successfully differentiated into osteoblasts and produced mineralized matrix.
Deposition analysis, bone/cartilage research: used for observing the deposition of calcium and other metals in bone tissue sections. Some universal reagent kits can also be used to detect the deposition of metal ions such as iron (FeCl) or cadmium (CdCl).
Cell staining operation:
1. Fixation: Fix the cell or tissue samples using fixatives such as paraformaldehyde.
2. Staining: Add alizarin red S staining solution and incubate at room temperature for a certain period of time (usually 5-10 minutes, depending on the amount of calcium deposition)
3. Washing: Wash thoroughly with distilled water or PBS to remove excess dye and avoid background interference. Note: The water temperature and washing time should not be too long to avoid dissolving the positive signal.
4. Observation: Observe directly under an optical microscope, and the calcium nodules appear orange-red.
Tissue staining operation (it is recommended to use 2% enhanced staining solution):
Remove wax until it reaches the water
1% Alizarin Red S staining for 2-5 minutes
Quick rinse with distilled water
Removable staining (such as hematoxylin, optional)
Dehydration, transparency, and mounting
