Alcian Blue Staining Solution

The Alcian blue staining kit is a special staining reagent commonly used in scientific research, primarily for observing acidic mucopolysaccharides and mucins in tissues or cells under a microscope, aiding researchers in identifying cartilage, gastrointestinal lesions, or tumor types.

Different pH values can selectively stain different types of acidic mucopolysaccharides:

pH 2.5: It can stain all acidic mucopolysaccharides (carboxylated and sulfated), primarily targeting substances with carboxyl groups, such as proteoglycans, hyaluronic acid, and epithelial acidic mucin. It is suitable for staining cartilage tissue and can also assess the degree of intestinal metaplasia in the gastrointestinal tract.

pH 1.0: Primarily stains sulfated mucopolysaccharides, commonly used in pathological studies of gastric cancer, lung cancer, etc., to help distinguish between mucinous sarcomas and liposarcomas

Operation of Alcian blue staining kit:

1. Fixation: Cell smears are commonly fixed with methanol for 5 minutes, while tissue sections require dewaxing and subsequent water washing

2. Staining: Add the staining solution dropwise and let it stain for 10-30 minutes (the specific time depends on the instructions provided in the reagent kit), then rinse with water.

3. Counterstaining: Commonly use carbol fuchsin for counterstaining for 5-10 minutes to make the cell nuclei appear red, facilitating comparative observation.

4. Mounting: After dehydration and clearing, mount the slide with neutral gum for microscopic examination

    Staining time: Generally 10–30 minutes, adjusted according to tissue type

    Storage: Store in the dark at room temperature. The staining solution can be reused multiple times, but it is recommended to replace it regularly to ensure the staining effect
